Characterization
Abstract
This year, the characterization of BBa_K199118 was been carried by our team. This part is composed by T7 promoter + RBS + mRFP, which produces red florescent protein. We originally use mRFP as our expression protein of our toehold switches. However, finding that it's hard to measure the OD value of mRFP in daily life, we finally change to use invertase as our expression protein.
We found out that different pH level might interfere with the reporter proteins, since different pH level could change the folding and structure. Thus, we want to know how pH value effect the performance of mRFP. We add mRFP we produce in vitro and different pH buffer includes pH 2, pH 5, pH 7, pH8, pH 10, pH12 into 384-well plate. Then we measure the excitation and emission spectrum and observe the different characteristics of them.
