Pilin Expression
Electrically conductive protein nanowire (e-PN) is the most fundamental and essential part of our project, which can generate the electrical power by utilizing moisture in the air. This kind of protein can be produced by Geobacter sulfurreducens, but it is an anaerobic bacteria which needs to be cultivated under strict conditions. Therefore, we decide only to use the pilA gene from Geobacter sulfurreducens and build the genetic pathway by ourselves.
The expression of e-PN in E. coli Top10 involves two parts: one part is the monomeric protein gene pilA(pilin gene) of e-PN, and the other part is the e-PN assembly system called TypeVI system, which comes from E. coli O157: H7 and MG1655.

RBS Mutation
We enhance the productivity of the protein nanowire by creating library of RBS hofB, hofM and ppdA in the Type IV pilin assembly system, so as to find the optimized combination of the three RBSs with varied strengthes.

Pilin Gene Mutation
We design 17 mutations of pilin gene for the purpose of adding more carboxylic groups into the nanowires and decreasing the diameter of them, so as to improve the output voltage of the protein film.
