This year, among the 41 parts designed, ten are basic parts and the other are composite ones. All of the parts are designed by ourselves, and most of them have been tested to insure their reliability. A series of composite parts have been designed this year, and most of them are useful for our project. And our favorite composite part is BBa_K2922009 , which contains the sequence of the protein Kil. Kil protein, which accumulates in the periplasmic space of the bacteria, can increase the periplasmic space and improved the membrane permeability of bacteria. Therefore, the protein inside the bacteria can secrete out of bacteria with the help from Kil protein. The use of proper intensity of the promoter can not only achieve the purpose of secretion but also prevent side effects of cell lysis and death. After a series of experiments, J23109 is determined as the most proper promoter.

This year, we have formulated a Part Collection related to secretion of intracellular proteins, which includes two major parts: one is secretion achieved with the help of YebF in the form of fusion proteins, the other is the expression of Kil protein by promoters of different intensities, thus changing cell permeability to different degrees and finally realizing secretion. For our Part Collection, the efficiency of three different intracellular proteins we utilized were well tested, recorded in detail and completely uploaded to the registry, which would be instructive in the field of design and characterization for many teams.
