This year, among the 103 parts designed, 44 of them are basic parts and the rest are composite parts. All of the parts are designed by ourselves, and most of them have been tested to insure their reliability. Among the 44 basic parts designed this year by ourselves, BBa_K3332011 is our favorite one, which comes from homo liver. The gene enables engineering bacteria to efficiently catalyze the reaction of reducing glyoxalic acid and consuming NADPH. Besides, many other basic parts have also been designed to construct our project.

A series of composite parts have been designed this year, and most of them are useful for our project. And our favorite composite part is BBa_K3332057 , which contains the sequence of the anchor protein—INPNC to anchor our favorite basic part—GRHPR.
The anchor protein is from a kind of autotransporter family which can be anchored to the membrane and work as a bacterial display system. The cleaved passenger domain remains adhered on the bacterial surface and it is conducive to keep the displayed exogenous proteins associated onto the cell. Thus, the functionalized bacteria can be recycled/re-used conveniently.
Throughout the season we’ve combined several surface display systems with GRHPR and verified the function of detecting NADPH outside the cell. After a series of experiments, INPNC is determined as the most suitable anchor protein for GRHPR.

This year, we have formulated a Part Collection related to surface display systems, which includes four kinds of anchor proteins: INPNC, AIDA, BrkA and Lpp-OmpA. The cleaved passenger domain remains adhered on the bacterial surface, which is conducive to keep the displayed exogenous proteins associated onto the cell, thus, the functionalized bacteria can be recycled/re-used conveniently.
Users can also combine the anchor proteins with different proteins to verify the function of the anchored proteins and treat it as a nice way to avoid the influence of components inside the cell. For our Part Collection, we’ve characterized three different anchored proteins combined with eGFP, GOX, GRHPR and iNap separately to test its efficacy and uploaded complete and detailed quantitative data to the registry, which is believed to be very instructive for the design and characterization of many teams.
