Basic Parts

1. BBa_K3581000: The alpha protein subunit making up the hagfish intermediate filaments which is registered in the iGEM registry by iGEM 14 UCC Ireland as BBa_K1518001. This part has a Acc65I cut site in front and AscI cut site behind, which allows chromoproteins to be inserted in the N or C terminus of the intermediate filaments for pre-dyeing purposes.




2. BBa_K3581001: The gamma protein subunit making up the hagfish intermediate filaments which is registered in the iGEM registry by iGEM 14 UCC Ireland as BBa_K1518000. This part has a Acc65I cut site in front and AscI cut site behind, which allows chromoproteins to be inserted in the N or C terminus of the intermediate filaments for pre-dyeing purposes.




3. BBa_K3581002: 6x His tag which is inserted in the C terminus of protein to allow purification and identification. The His tag can be identified and purified using nickel (II) ions in affinity purification columns or anti His tag antibodies. This part has a TEV protease cut site in front of the His tag sequence which can be used to cleave out the His tag after the protein has been purified. The sequence is originally designed and registered by iGEM 13 BGU Israel.




4. BBa_K3581003: HlyA signal peptide used to target protein secretion via Type 1 secretion pathway by attaching signal peptide to C terminus. This allows the protein to be secreted and transported from cytoplasm to extracellular space. This part is flanked by HindIII cut sites because it allows the signal peptide to be cleaved out so the secretion method can be changed from extracellular to inclusion bodies. The sequence is originally designed and registered by iGEM 09 Utah State as BBa_K208006.




5. BBa_K3581014: TorA signal peptide is used to target protein secretion by having fully or partially folded proteins secreted to the periplasm. This part is flanked by HindIII cut sites because it allows the signal peptide to be cleaved out so the secretion method can be changed from extracellular to inclusion bodies. The sequence is originally designed and registered by iGEM 19 Calgary as BBa_K3114005.




6. BBa_K3581004: aeBlue is a blue chromoprotein designed by iGEM 12 Uppsala-Sweden and registered as BBa_K864401. It is obtained from the basal disk of a beadlet anemone Actinia equina. The protein has an absorption maximum at 597nm and a deep blue color. The part is flanked by Acc65I and AscI cut sites, which allows it to bind to the N or C terminus of the protein depending on our experimental goal.




7. BBa_K3581005: eforRed is a red chromoprotein designed by iGEM 11 Uppsala-Sweden and registered as BBa_K592012. The protein naturally exhibits red/pink color when expressed. The part is flanked by Acc65I and AscI cut sites, which allows it to bind to the N or C terminus of the protein depending on our experimental goal.




8. BBa_K3581013: amilGFP is a yellow chromoprotein designed by iGEM 11 Uppsala-Sweden and registered as BBa_K592010. The protein exhibits yellow colour when expressed and it can be found in the coral Acropora millepora. The part is flanked by Acc65I and AscI cut sites, which allows it to bind to the N or C terminus of the protein depending on our experimental goal.
