Our main job is to design the genetic circuits that allow Y. lipolytica to use starch and produce biofuel. We have constructed six basic parts, including EXP promoter, AnGlu (glucoamylase), AnAmyA (alpha-amylase), XpR2t terminator, Leu marker. Three composite parts are assembled step by step by Type IIS Assembly Methods, including EXP-AnGlu-XpR2t-Leu (BBa_K3578010) and EXP-AnAmyA-XpR2t-Leu (BBa_K3578011), EXP-AnGlu-XpR2t-EXP-AnAmyA-XpR2t-Leu (BBa_K3578012). This year we constructed EXP-AnGlu-XpR2t-Leu and EXP-AnAmyA-XpR2t-Leu, which were transferred to Y. lipolytica to verify the starch utilization efficiency and lipid production.
