Partnership

We and XHD-ShanDong-China, as a brother team, have a very deep cooperation in the whole season from learning the basic knowledge of synthetic biology, to adjusting the direction of project design, to the implementation of project experiments, including result analysis. All of these have benefited each other greatly and played an important role in the successful implementation of our project.

1. Basic knowledge learning and sharing / Weekly project meeting

Our team was successfully established in May, and began to learn the basic course of synthetic biology. As wiki is an important part of the whole project, our team members sorted out the wiki edit materials and shared it with XHD-ShanDong-China team members, which helped them a lot. In addition, as we are under the guidance of the same PI teacher, we have weekly project study and Discussion online, which greatly promotes our project cooperation.

2. Application for public wechat

They encountered difficulties when editing the official account number. Our captain Hu Keer helped us deal with this problem.

3. Project design

At the beginning, when we designed our project, we didn't consider including both contribution and improvement into the complete scheme of the project. Because referring to the past experience, the parts of contribution and improvement parts can be independent of project objectives. However, after our discussion and analysis, we all feel that it is better to find parts related to our project to contribute and improve. On the one hand, it can enrich our project experiments and make full use of the results. Secondly, the project will be more integrated into one.

4. Technical cooperation of project experiment

We found our two teams needed similar experimental methods , such as plasmid recombination, bacterial culture, growth curve drawing, molecular cloning and so on. In these aspects, we shared the common experimental operation protocol. we sent students to share the work of finding the experimental protocol, and then summarized and shared them.

Especially in determining the methods of plasmid recombination, we know that there are digestion link method and homologous recombination method. Finally, after consulting the literature and discussing with teachers, we choose homologous recombination method to insert DNA fragment, which can insert the specified sequence at any position. The experiment was carried out smoothly.

In the modeling part, we share some references, including but not limited to FFL, spatial dynamics, enzyme kinetics, etc.

5. Troubleshoot in project implementation

1) Video editing

As their team did not have students who are good at editing videos, our team member Yang Mutian gave them great support and help in promoting the background music synthesis and animation production of the video.

2) WiKi editing

As we all know, Wiki editing is very important. When they try to edit the team home page, they accidentally delete the top bar, which makes it impossible for them to access the edit page from this tool. Because this page is the home page, they can't give up. They asked us for help. our team's member DingDing Chen helped XHD-ShanDong-China successfully solve the wiki website editing technical problems.
