Partnership
We and XHD-Wuhan-China, as a brother team, have a very deep cooperation in the whole season from learning the basic knowledge of synthetic biology, to adjusting the direction of project design, to the implementation of project experiments, including result analysis. All of these have benefited each other greatly and played an important role in the successful implementation of our project.

Our team was set up later than XHD-Wuhan-China and there are a lot of self-taught courses need to study, especially wiki edit. XHD-Wuhan-China friendly provided us with Wiki production materials . As we are under the guidance of the same PI teacher, we will study and discuss online every week, from basic knowledge of synthetic biology to excellent project learning, to the progress and communication of each other's projects. We will exchange and discuss on the Internet regularly. Although the epidemic has hindered our offline gathering, online communication and learning can greatly promote our project implementation.
1) At first, we planned to look for literature for the contribution part, but the students of XHD-Wuhan-China suggested that if we could come to Wuhan to do the experiment, we should try to do the experiment, and it would be better to have objective and detailed results. Finally, in the aspect of experimental implementation, it is they who help us complete some basic experiments, which makes our later experiment progress more smoothly.
2) In addition, our project, from the selection of contribution parts, to the genes of the project, and then to the selection of improved parts, through discussion with XHD-Wuhan-China members, all the functions of the parts can be closely related, reflecting the integrity of our project design ideas in engineering. Instead of just conducting experiments to obtain a certain standard, we spent a certain amount of time screening parts, because our project experimental plan is to determine the pH value, so we selected acid sensitive promoters in the screening of contribution parts, and selected parts related to our project in the process of improvement.
We found that both of our teams needed to recombine plasmids and culture bacteria to draw growth curves. The methods of plasmid recombination include Digestionlink or homologous recombination. Finally, after consulting literature and discussing with teachers, we chose homologous recombination method to insert DNA fragment, which can insert the designated sequence at any position. The experiment process can be carried out smoothly. Finally, we share a set of homologous recombination experiments. In the modeling part, we shared some references, including but not limited to FFL, spatial dynamics, enzyme kinetics and so on.
Because our team did not have students who are good at editing videos, we invited Yang Mutian from XHD-Wuhan-China to assist us in completing this work. In the later supplementary experiment, he also made a lot of contributions to our team, so we invited him to become a member of our team.
Our team encountered a very difficult problem when we first started wiki editing. When we tried to edit the main page of the team, we accidentally deleted the topbar, which made it impossible for us to enter the editing page of the web page from this tool. Because this page is the main page, it can't be abandoned. We tried many ways to solve this problem, but we didn't make progress. We asked for the help of XHD-Wuhan-China, they found that if we replaced the name of the team in their homepage with our name, we could directly enter the editing page of our team. So we used this method to retrieve the editing page of our team's homepage, and restored the code. Finally, we restored our home page to normal.
We encountered difficulties when editors applied for the WeChat official account. Their Team leader Hu Keer helped us deal with this problem.
