Overview

In order to increase the production of exopolysaccharides (EPSs), we altered the levels of enzymes in the central metabolism. Through a large number of literature investigations, we selected and over expressed two key enzymes, galU and pgmA.Then six composite parts have been constructed containing different combinations of extracellular polysaccharide synthesis gene galU and pgmA, to achieve maximal yield of extracellular polysaccharide, which is the key component of biological soil crusts for desertification control. Parts number ranges from BBa_K3331009 to BBa_K3331014.

In terms of biosafety, we have constructed a suicide switch controlled by pBAD. We found two sets of arabinose operon sequences, which were derived from Escherichia coli and Bacillus subtilis respectively. We constructed verification plasmids to verify their performance.
