Project Description In fact, the COVID-19 epidemic has greatly influenced the design of our project, and we finally completed the design of our project after consulting many professors and doctors. Achieve
Contribution In the experiment, we characterized the related functions of BBa_K1633008 and BBa_K2796028. And we designed the new part of the functional comparison, to contribute to the future of the team. Engineering Success BBa_K3335004 and BBa_K3335005 are siRNA targeting PD-L1 and CD47 was designed, and siRNA was designed in a completely different way.We characterize it at the cellular level and verify that it can be expressed normally. BBa_K3335000(KIBRA) as an adaptor-like protein that stabilizes Rab27a, which in turn controls exosome secretion. In our experiment, we used KIBRA to promote cell release of exosomes containing siRNA, thus increasing the efficiency of cell production and release of exosomes and improving the killing of tumor cells. Collaboration We have good communication with iGEM teams. We hold GenScript Bioethics meeting with ZJU-China, NJMU-China, NJTech_China, ShanghaiTech_China. Meanwhile, we often have good communication and cooperation with high school teams, Korea-HS and Worldshaper——Wuhan to make Suggestions on the implementation of their projects. At the same time, we helped ShanghaiTech_China complete the content of 3D printing. Proposed Implementation We investigated the global epidemic of COVID-19, focused on the treatment problems faced by cancer patients during the epidemic, and got in touch with Master Wang Tingting to understand the status of patients. Therefore, we put forward our project concept. Meanwhile, we interviewed Dr. Liu Rui and Dr. Wang Rui and got the improvement of the project. Integrated Human Practices Our project is closely integrated with society. We started with a thesis about cancer patients during the epidemic. Through the paper and the information we have collected, we understood the difficult situation of cancer patients during the epidemic. We decided to do something to help them. Then we interviewed some cancer doctors and professors who research cancer therapy. According to their suggestions, we added KIBRA protein and Dox switch, which made our project safer and reliable. Our goal is to create a safe and effective treatment. However, scientific perfection does not mean social acceptance. Therefore, we launched investigations into the acceptance and ethical issues of modified organs. Through interview and questionnaire, we proved that if our project is safe enough, it can be recognized by the society. Improvement of an Existing Part In order to design a better KRAS siRNA, we optimized the siRNA sequence provided by BBa_K1942000. A new siRNA part BBa_K3335003 was designed and its expression and function were characterized. Its ability to knock down mRNA levels was significantly better than that of BBa_K1942000. Project Modeling Our project uses the Tetracycline on(Tet-On) system, which can only be expressed through Tetracycline. In order to select the appropriate tetracycline concentration for subsequent experiments to turn on the switch. We modeling the changes in blood levels in mice after oral administration of tetracycline and the effects of different concentrations of tetracycline on the intensity of TteO-CMV expression. Through the analysis of modeling Result, we can choose different concentrations of tetracycline at different times for personalized drug delivery in the future. Achieve
Proof of concept Through literature review and experimental data support, we proved that the design is effective and can work. The control system can effectively and safely ensure the switch on and off. RNAi system can ensure that siRNA can effectively knock down the expression of related mRNA. The targeting system ensures the targeting of exosome delivery. Exosome release system ensures efficient exosome release. These combinations ensure the availability and stability of the project. Partnership In the past year, we have opened a close and full cooperation with Zju-China.Discuss and complete the project design and improve each other in the early stage of the project.Help each other with Eastern China meet-ups and Opening Weekend activities.In the middle of the project, we discuss the issue of project safety together, collect the public's consideration of safety, hold a meeting to discuss the issue, and go to GeneChem to discuss the possibility of industrialization. Science Communication In the Education part, our team created a colorful internal microcosm for people without biological backgrounds to understand. We took the advantage of the cartoon to bring people into the world of synthetic biology, and on the media platform we have attracted a lot of followers. Apart from this, we gave a lecture to the students from Jinling High School and attended the publicity activity for university associations in Nanjing university. The former was recognized by high school students through project introduction and Q&A, while the latter obtained feedback from college students through soliciting their attitudes towards our cancer treatment methods. What is more, we also interviewed teachers and doctors on Opening Weekend to optimize our design solutions and better publicize our program. In conclusion, we adopted appropriate methods to promote synthetic biology and our project to populations in different fields. Achieve
