Wet Lab Notebook
Our journey in the lab started in December 2019. Back then, the wet lab team's undergraduate members were trained under the guidance of the MSc students and our Instructor. The training's purpose was to familiarize with basic experimental protocols performed in the laboratory, such as DNA extraction of whole blood samples using an extraction kit, PCR, and electrophoresis. An essential part of the training was the biosafety instructions that ensured the smooth conduct of the experiments. During the training, the mock-tests were performed using water instead of real reagents, multiple times until they were fully prepared to successfully and safely conduct the experiments.
Meanwhile, we conducted a literature search for the targeted gene, SLCO1B1, the polymorphism rs41049056, and the referred drug correlations. Also, the designing of primers to be used in the AS-PSR was carried out.
In January 2020, the first experiments were performed to standardize our experiments' protocol, and we continued as presented in our notebook below.
Due to the COVID-19 pandemic, the frequency of our experiments was reduced during March and April. Still, fortunately, the President of the Department of Pharmacy of the University of Patras and our Principal Investigator, Prof. Patrinos, managed to ensure our access to the laboratory, respecting all the safety measures to prevent the spread of the virus.
