After we decided to use toggle switch as the principle of our circuit, and use lac as a molecule in our circuit, we used these keywords to find the most suitable system for our yeast, and we found the article describing a hybrid promoter: nmt::lacO promoter.
We organized the information in the research and add some information to BBa_C0012.

Description

nmt::lacO promoter(hybrid promoter mentioned in the following passage) is an IPTG‐inducible system which is the derivative of the Sz. pombe nmt promoter. In this system, endogenous nmt promoter in combines with the heterologous lacO–lacI system,  and allows expression in a short and defined window.


The hybrid promoter described here has several advantages:
1.Induction is guaranteed by addition of a simple agent (IPTG) with no or minimal effect on cell metabolism
2.The system displays a relatively wide and dynamic range of activity
3.Full induction of the system can be achieved within a short period of time
4.The system can easily be switched off by the addition of thiamine
5.The inducer, IPTG is authenticated safe.

Information we added

1.If a chimeric lac operator site (lacO) is positioned appropriately downstream of the TATA‐box, gene expression from a nmt::lacO promoter (hybrid promoter) can be greatly repressed by lacI inhibitor.

2.Insertion of the operator sequence that contains the lac operator into the nmt promoter does not interfere with the function of the promoter.

3.The addition of IPTG to the medium could relieve the repression to some extent, but it is not fully reversible.
