We participated in the East China exchange meeting. Among them, Mr. Zhu Tianze from Beijing Zino Technology gave us suggestions, and we also participated in the drift bottle project.

Participated in the online exchange meeting in East China on the 18th of this month. In the meeting we explained our experimental design and project prospects in detail. We communicated with Northeast Forestry University on gene circuit design and project planning, social practice, etc.

August 2020

From 3th to 10st of this month, Together with THU, PKU, CAU, and BNU, we launched a one-week open educational popularization of synthetic biology online.

On 26th, we discussed the design rationality of the integrated fermentation system with BNU online.

September 5 to September 13, 2020

After the epidemic went back to school, I came to the school and asked the instructor. I was guided by PI in the experimental design and provided us with an experimental site.

We interviewed Academician Lin Zhanxi and discussed the problems and prospects of Juncao fermentation to produce fiber bioethanol

We held popular science activities on synthetic biology in the school, and the safety issues brought by synthetic biology

September 14 to September 18, 2020

We officially started our experiment, we began to screen cellulolytic bacteria from the soil and successfully isolated a single colony

We studied and designed our primers and purchased the reagents and kits needed for future experiments

On 16th, we discussed the Problems encountered during the experiment with BNU online.

September 18 to September 22, 2020

We successfully constructed the B3 non-gate fragment into the Pet28b plasmid.

September 22 to October 1, 2020

We successfully transformed the recombinant plasmid into BL21 competent cells and got the correct transformants.

We continue to purify the isolated single colony and save it for backup.

October 2 to October 9, 2020

We interviewed teacher Liu Bin, and he suggested that we use Beauveria bassiana against our degrading enzyme system and pay attention to the best harvest period for Juncao suitable for fermentation.

We compare the decomposed cellulolytic bacteria and white pulp bacteria in various aspects, and use the DNS method to test and verify the enzyme activity of our cellulolytic bacteria.

We measured the growth curve of BL21.

October 10 to October 19, 2020

We continue to purify the separated cellulolytic single colony,

We used the hemicellulose kit purchased by Solarbio to determine the content of hemicellulose in the grass powder.

We successfully constructed RFP and GFP into the PET28 plasmid, and transformed into DH5α, and saw green fluorescence.

We interviewed Jingfeng Technology Co., Ltd. and Xinda Knitting Co., Ltd. We visited their textile factories, and the staff discussed with us the economic problems of using Juncao fermentation to produce cellulose.

We communicated online with Jilin University and learned from each other’s work system
